DNA methylation is widely recognised as a fundamental epigenetic mechanism, both essential for normal embryonic development as well as abnormal in a range of genetic diseases. CpG Methylation is the most widely investigated epigenetic modification in humans. The appreciation of its fundamental role in development, gene silencing and genomic stability has subjected the changes in DNA methylation status to ever-increasing scrutiny over the last 20 years. Pyrosequencing is a fast, simple and quantitative method for analysis of CpG methylation in multiple sites in a single assay, eliminating the reliance on restriction sites, large quantities of DNA and gels.
